This feature is included in every MTH PS2 subway set, including the R21 which was MTH's first PS2 subway set. This feature is available both under DCS and when running conventionally.
I believe that Lionel has a feature that calls out individual stations manually, however, it does not allow the operator to set up a pre-programmed subway route. I also don't know if his feature requires that the subway be running under Legacy, or not. Hopefully, someone more knowledgeable than I will be able to provide more information regarding the Lionel subway. One thing you have to watch with the R21 is that there were two versions One of them has PS2 and can be used as Barry states and the other version only has locosounds.

And to add to that,1 had a heat reflective tape on the inside of the roof and the other didnt.And as a result would bubble up on the roof when ran for a long time.

The Lionel R-27 Railsounds is for the QT train (the route on the signboards) from Ditmars Blvd in Queens to Stillwell Ave in Coney Island, Brooklyn.
